The 2000–01 Conference League Cup, known as the Nationwide Variety Club Trophy for sponsorship reasons, was a football tournament for clubs competing in that season's Football Conference. It was the 22nd edition of the tournament, and was thought to be the last after being axed, but it would briefly be reformed in 2004–05. It began withe the first round in September 2000, and concluded with the final on 7 May 2001.

Doncaster Rovers were the defending champions, but they were eliminated by Southport in the second round on 17 October 2000.
